New prostate cancer epigenetic markers are found by researchers
New epigenetic biomarkers have been found by researchers at the Garvan Institute of Medical Research to predict more aggressive forms of prostate cancer. Published in the journal Clinical and Translational Medicine is the latest study. The biomarkers could aid doctors in creating a more effective treatment strategy and be used in conjunction with conventional clinical methods to forecast if a man will eventually acquire a more deadly and metastatic form of the disease.
According to Professor Susan Clark, Head of the Epigenetic Research lab at Garvan and lead researcher of the study. , “men with prostate cancer need to have more personalised treatments guided by the nature of their tumours, and they can’t get that without new biomarkers that can better predict the risk of developing the lethal form of the disease.”
The second most prevalent cancer among men to be diagnosed is prostate cancer. About half of all men with cancer will eventually acquire metastatic cancer after their initial diagnosis. However, a tiny number of men experience a deadly, metastatic form of the disease much sooner after diagnosis. Metastasis typically takes 15 or more years to develop. Clinicians could start more aggressive therapies earlier by identifying patients who might go on to acquire this form of prostate cancer in the early stages.
One of the longest and most thorough molecular studies on the development of prostate cancer is this one. It is challenging to research the disease’s biology because of how slowly it progresses.
The researchers were able to analyse samples from 185 men who had their prostates removed after being diagnosed with prostate cancer in the 1990s and 2000s because to the bank of biopsies that has been kept at Garvan and St. Vincent’s Hospital for the past 20 years. The researchers then kept tabs on how many men survived and how many passed away from the illness, some more than 15 years later.
The scientists examined their genomes and discovered 1420 sites that were unique to prostate cancer and where they could observe epigenetic alterations, or DNA methylation marks. Without changing the DNA sequence of a gene way a mutation does, the methylation process can increase or decrease a gene’s activity
Of those locations, 18 genes were further investigated, with the CACNA2D4 gene—which is involved in calcium channel regulation—standing up as a significant biomarker.
We urgently need to comprehend how the methylation process may decrease the gene’s function because there is very little information available about this gene and it is not frequently profiled, according to Dr. Ruth Pidsley, the study’s first author.
The group has made the detailed epigenome sequencing data accessible for use by other researchers in their work on prostate cancer.
The results of the epigenome study revealed variations between individuals with the lethal and non-lethal types of prostate cancer, and the biomarkers also increased the prognostic accuracy of currently used clinical tools.
The new discoveries offer hope for a technique to treat cancer in a more individualised manner.
As the clinical lead on the trial and an oncologist at Garvan, Professor Lisa Horvath says, “What you really want to know on the day a patient is diagnosed is who has the potential for fatal prostate cancer and who does not, since it will impact the way you treat the cancer.”
These epigenetic indicators may be able to identify who has fatal prostate cancer and who does not, according to the researcher.
The investigation will be expanded, and the following steps will establish whether the biomarkers can initially be found in blood samples.
